Hm, I hadn't thought about that life debt in relation to the Tower scene, but now I realise it was an additional reason why he had to kill Dumbledore. The way I see it, he had no hope of saving Dumbledore and had to choose between a glorious (but useless) Gryffindorish death for himself, Harry and Darco, with the prospect of many other students getting killed by the Death Eaters afterwards, or Slytherinly saving what could be saved. The Unbreakable Vow to protect Draco coupled with the life debt to James really made it no contest, no matter how reluctant he was to off Dumbledore.
